Disease Model Phenotype Type II Diabetes ob/ob or db/db (genetic model) ob/ob is a mutant mouse that eats excessively due to mutations in the gene responsible for the production of leptin and becomes profoundly obese. Mutant mice gain weight rapidly throughout their lives, reaching a weight three times that of unaffected mice. ob/ob mice develop high blood sugar, despite an enlargement of the pancreatic islets and increased levels of insulin. db/db mice express mutations in leptin receptor that lead to decreased insulin receptor sensitivity and subsequent increased levels of blood glucose, decreased β-cell function, increased obesity and elevated HBA1c levels.
Type I Diabetes NOD (genetic model) The NOD/ShiLtJ strain (commonly called NOD) is a polygenic model for autoimmune type 1 diabetes. Diabetes in NOD mice is characterized by hyperglycemia and insulitis, a leukocytic infiltration of the pancreatic islets. Marked decreases in pancreatic insulin content occur in females at about 12 weeks of age and several weeks later in males
obesity High fat diet-induced C57BL/6 is among the most sensitive to this diet-induced obesity (DIO). C57BL/6-DIO mice, particularly males, develop severe obesity, increased glucose intolerance, moderate insulin resistance and elevated glucose levels, but they typically do not develop basal hyperglycemia (from JAX).
Dysregulated metabolism and insulin resistant IfitmDel (genetic model) The IfitmDel mouse lacks all five of the Ifitm genes via LoxP deletion. The IfitmDel animals exhibit a steady and significantly enhanced weight gain relative to wild-type controls. The increased weight corresponds with elevated fat mass, and in tolerance tests they are hyporesponsive to insulin but respond normally to glucose. Both young (4 mo) and older (12 mo) IfitmDel mice have enhanced levels of serum leptin.
